Uncover Signaling-Regulated Transcription Factors in Hematopoiesis | Master's Directed Studies

I analyzed CITE-seq data, a combination of single-cell RNA sequencing and protein expression, from hematopoietic stem cells using Python. My goal is to uncover the key players – signaling-regulated transcription factors specific to each cell type – that control how these cells transform into different blood cells. To achieve this, I preprocessed the data, grouped similar cells into metacells, and then applied a powerful algorithm called SPaRTAN (Single-cell Proteomic and RNA based Transcription factor Activity Network). This revealed the crucial transcription factors that orchestrate this dynamic process at the single-cell level, offering exciting insights into blood cell development and potential therapeutic targets for blood disorders.
